flexTEC systems - our basic Toggle systems:


The flexTEC systems are the basic implementation of the ToggleTec vector lay-out. These are ready-to-use with the appropriately designed gene cassettes.

ATG:biosynthetics GmbH has developed a vector-based gene assembly system with standardized modules that adhere to a uniform design logic. This system allows easy modification, exchange, addition, removal and regeneration of components or higher-order gene assemblies. This system has been de-signed with the idea of an industrial standard in mind and has successfully been implemented for multi-gene assemblies in metabolic engineering of bacteria.

tANCHOR:

tANCHOR technology: The new technology for exposing proteins on cell surfaces. The system is well suitable for peptide/ epitope/ protein display on the surface of mammalian cells applying two different members of the tetraspanine family of proteins, CD63 (HEK293T) and CD82 (HeLa) each expressed in various cell lines a little better. In addition the tANCHOR vectors are available with terminal mCHERRY fluorescence protein.

flexColi Basic, flexColi Turbo

flexColi in its basic implementation is a system that lets you select from a variety of A-Box and S-Box functions (refer to table). The turbo versions are for high expression levels.

The flexColi starter Kit includes T7 and lac - promoter variants, you may choose other advanced configurations as well. The flexColi transfer vectors consist of 2 acceptors and 4 donors:

flexBACTurbo

flexBACTurbo in its basic implementation is a system that lets you select from two A-Box (with polh and p10 promoters, with/without Tn7 attachment sites) and three S-Box (KanR, GmR, CmR) functions you require (refer to table).

In the basic (CLASSIC) set, TogBac transfer vectors come with 2 acceptors and 4 donors:

Genes cloned into the transfer vectors can the be recombined into multi-gene cassette assemblies by Cre-loxP recombination or via toggle concatenation.

Cre-Decre-cascade Sample Cre-DeCre cascade of assembling and disassembling multi-gene (cassette) constructs. The red circles mark the loxP recombination signal(s). In this example, donors have an R6K gamma origin of replication but they may as well possess a ColE1 ori.

All vectors host a split ori which allows conversion from a medium/high-copy ori (ColE1) into an R6K-gamma conditional ori by a simple restriction-ligation reaction.
